Weak flexible polyelectrolytes carry ionizable groups whose charge state fluctuates through acid-base equilibria (charge regulation), and these charge fluctuations (CF), quantified by the binding capacitance, are known from Monte Carlo simulation to fold the chain more than a charge-averaged treatment predicts and to shape its low-force stretching response. However, no first-principles mechano-statistical theory exists that explains these effects: in the 2019 simulation study by Blanco et al., the authors state that a theory analogous to the Kirkwood-Shumaker theory of fluctuation-induced attraction between protein molecules, but acting between different regions of the same chain, would be desirable to understand whether and how binding capacitance and CF generate effective intrachain attractive contributions and quantitatively modify conformational statistics. This problem asks for that theory: a derivation from a microscopic model of a protonating flexible chain that clarifies whether charge fluctuations yield a fluctuation-induced intrachain attraction and quantitatively predicts the gauche-state probability, persistence length, and pH-dependent Pincus stretching exponent.

Background

Weak polyelectrolytes are polymers bearing ionizable groups (for example amines or carboxylates) whose charge is set by acid-base equilibria, so the average macromolecular charge responds to pH, ionic strength, and other perturbations — a behavior called charge regulation (CR). In a constant-pH (semi-grand canonical) description each site's protonation fluctuates, and the strength of these charge fluctuations (CF) is measured by the binding capacitance, the response of the average degree of protonation θ\theta to pH. CF matter because correlated fluctuations of the ionization states of two charge-regulating objects can weaken, or even make attractive, their electrostatic interaction: the classic Kirkwood-Shumaker theory shows that two like-charged protein molecules experience, besides the mean-charge repulsion, an attractive force proportional to the covariance of their proton charge fluctuations. Whether an analogous fluctuation-induced attraction acts within a single flexible chain — between different regions of the same polyelectrolyte — is much less understood.

Blanco, Madurga, Narambuena, Mas and Garcés (Polymers 2019) studied this by Monte Carlo simulation of a 'minimal' model of a flexible weak polyelectrolyte: proton binding to sites, trans/gauche rotational-isomeric bond states (gauche being the compact local conformation), bond stretching and bending, and Debye-Hückel-screened electrostatics (site-site electrostatic interactions whose range is reduced by the dissolved salt, decaying over a length set by the ionic strength), with steric excluded volume (a hard-core repulsion forbidding overlap of distant chain segments). Comparing constant-pH simulations (semi-grand canonical Monte Carlo, SGCMC), in which each site's charge fluctuates through proton exchange with the solution, with constant-charge Monte Carlo simulations (ccMC) that freeze every site at its average charge, they found that CF raise the gauche-state probability and lower the persistence length (the distance over which the chain direction stays correlated), i.e. the chain folds more than a charge-averaged treatment predicts, most strongly at intermediate θ\theta where the binding capacitance is maximal. Under stretching, in the low-force regime the extension follows the Pincus scaling law Lz∝F1/ν−1L_z \propto F^{1/\nu-1}, where FF is the applied force and ν\nu the Pincus exponent: ν=3/5\nu=3/5 is expected for a strongly charged self-avoiding chain (swollen by repulsion between its segments) and ν=1/2\nu=1/2 for a neutral phantom chain (an ideal chain whose segments can pass through one another, so it has unperturbed-coil statistics). The simulations show a pH-dependent crossover between these values, but at high ionic strength (1 M) the value 3/5 is not reached even when the chain is fully charged (θ=1\theta=1). The authors state that a mechano-statistical theory from first principles, similar to Kirkwood theory for interacting macromolecules, would be desirable to understand the fluctuation-induced folding, and that no such theory exists. Available analytical tools do not fill the gap: the Site Binding-Rotational Isomeric State (SBRIS) model — a transfer-matrix treatment, in which the chain partition function is built by multiplying matrices that couple the ionization and conformational state of each bond — treats conformation-ionization coupling exactly but only for short-range interactions, and mean-field single-chain theories of ionization do not produce a Kirkwood-Shumaker-type decomposition of the intrachain interaction into mean-charge and fluctuation contributions.

Problem Statement

Derive a first-principles mechano-statistical theory for a single flexible weak polyelectrolyte in solution whose ionizable sites undergo protonation equilibria. Starting from a stated microscopic model — proton binding to sites, trans/gauche rotational-isomeric conformational states, and screened electrostatic site-site interactions — obtain the effective interaction between different regions of the same chain, clarifying whether and how charge fluctuations, quantified through the binding capacitance or site-charge correlation functions, generate a Kirkwood-Shumaker-like attractive contribution in addition to the mean-charge repulsion. The theory must quantitatively predict how charge regulation and charge fluctuations modify (a) the conformational statistics of the free chain — the gauche-state probability and the persistence length as functions of the average degree of protonation θ\theta and the ionic strength — and (b) the low-force mechanical response — the pH-dependent Pincus exponent ν\nu in Lz∝F1/ν−1L_z \propto F^{1/\nu-1} and its crossover between the phantom-chain value 1/2 and the strong-charge value 3/5. The theory must be compared quantitatively against the published constant-pH Monte Carlo results of Blanco et al. (Polymers 2019, 11, 1962), reproducing the reported trends to within the accuracy of those simulations.
